چکیده

Monolayers of cysteine linked peptide nucleic acid (PNA) assembled on gold electrodes were investigated for the sensing of DNA recognition. The monolayer was characterized by cyclic voltammetry (CV). The electron transfer through the monolayers was investigated using electrochemical impedance spectroscopy (EIS) in the presence of target DNA and redox marker ions [Fe(CN)6] 3 /4 as hybridization indicator. The EIS spectra in the form of Nyquist plots were analyzed with a Randles circuit, and the electron transfer resistance, Rct, was used to monitor the sensing of the target DNA. The results showed that without any modification to the target DNA, the complementary and the mismatch can be discriminated effectively at PNA self assembled monolayers (SAMs). Therefore, this work can provide another view on using PNA SAMs as a probe candidate for label-free detection of DNA recognition. 2007 Elsevier B.V. All rights reserved.

A new Schiff base 2-aminothiophenol furfural self assembled monolayer (SAM) has been fabricated on a bare gold electrode as a novel sensor for determination of dopamine. Electrochemical impedance spectroscopywas utilized to investigate the properties of the Au 2-aminothiophenol furfural self assembled monolayermodified electrode (Au ATF SAM-ME) using the [Fe(CN)6]3-/4- redox couple. The electro...

Label-Free Potentiometry for Detecting DNA Hybridization Using Peptide Nucleic Acid and DNA Probes

Peptide nucleic acid (PNA) has outstanding affinity over DNA for complementary nucleic acid sequences by forming a PNA-DNA heterodimer upon hybridization via Watson-Crick base-pairing. To verify whether PNA probes on an electrode surface enhance sensitivity for potentiometric DNA detection or not, we conducted a comparative study on the hybridization of PNA and DNA probes on the surface of a 10...

DNA Hybridization Sensors Based on Electrochemical Impedance Spectroscopy as a Detection Tool

Recent advances in label free DNA hybridization sensors employing electrochemical impedance spectroscopy (EIS) as a detection tool are reviewed. These sensors are based on the modulation of the blocking ability of an electrode modified with a probe DNA by an analyte, i.e., target DNA. The probe DNA is immobilized on a self-assembled monolayer, a conducting polymer film, or a layer of nanostruct...

A Label-Free Electronic Biosensor for Detection of Bone Turnover Markers

This paper describes the development of a biosensor based on label-free immunosensing for the detection of the C-terminal telopeptide bone turnover marker from type-1 collagen. A self-assembled monolayer (SAM) of dithiodipropionic acid was deposited on a gold electrode. Then streptavidin and biotinylated anti-human C-terminal telopeptide antibody were successively conjugated on the self-assembl...
